Feather Sample Collection Protocol¶
Feather samples are the most commonly submitted sample type to SENO's avian genetics laboratory. When collected correctly, freshly plucked feathers provide reliable DNA for sexing, genetic disease screening, and species identification. This protocol covers proper plucking technique, quality control, species-specific guidance, and packaging standards.
Why Freshly Plucked Feathers Are Essential¶
DNA quality is the single most important factor for accurate testing. Feathers must be freshly plucked — the DNA in feather samples comes exclusively from the follicle cells at the base of the feather shaft. Shed feathers found on the cage floor have desiccated follicles and will not yield viable DNA for PCR amplification.

Step-by-Step Collection Procedure¶
Step 1: Restrain the Bird¶
Restrain the bird gently but securely. For small birds, use a towel or cupped hands. For larger birds (parrots, macaws), a second person may be needed to stabilize the head and body.
Step 2: Identify the Collection Site¶
Select feathers from the chest or flank region. These feathers have the best follicle development and provide the highest DNA yield. Avoid wing and tail feathers — they are more painful to remove and the follicles may be damaged.

Step 3: Pluck the Feather¶
- Grasp the feather close to the skin using thumb and forefinger
- Pull firmly and quickly in the direction of feather growth
- Inspect the follicle — a small tissue bulb should be visible at the feather base
- If the bulb is absent or appears dry, the feather was shed — discard and collect another

Step 5: Place Feathers into Envelope¶
- Open the paper envelope and drop the feathers directly inside
- Do not touch the feather base with fingers — oils from skin can contaminate the sample
- Fold the envelope flap but do not seal until ready to ship (air circulation prevents mold)
- Label the envelope immediately with bird ID, collection date, species, and owner name
Step 6: Verify Sample Quality¶
Before finishing the collection, confirm the following quality criteria:

- [ ] Follicle bulbs visible on all feathers
- [ ] No visible blood, fecal material, or dirt on feather shafts
- [ ] Feathers are dry and intact

Species-Specific Guidance¶
Psittacines (Parrots, Macaws, Cockatoos)¶
Large psittacines have strong, well-developed chest feathers. Collect 2–3 feathers from the upper chest. For macaws, the feathers are thick — ensure the follicle bulb is intact after plucking.
Passerines (Finches, Canaries, Sparrows)¶
These small birds have delicate skin and fine feathers. Use sterile forceps for better control. Collect 5–7 breast feathers. Watch for signs of stress and stop if the bird shows distress.
Columbiformes (Pigeons, Doves)¶
Pigeons have loosely attached chest feathers that pluck easily. Collect 3–5 feathers. Pigeon feathers may have less visible follicle tissue — inspect closely and consider a blood or swab sample if follicle quality is poor.
Galliformes (Chickens, Turkeys, Quail)¶
These birds have dense feather coverage. Collect 3–5 feathers from the breast or flank. Avoid blood feathers (newly forming feathers with a blood-filled shaft).
Raptors (Hawks, Falcons, Owls)¶
Raptor feathers are tightly attached and require more force to pluck. Collect 2–3 chest feathers. For sensitive species, consider a blood or swab sample as an alternative.
Waterfowl (Ducks, Geese, Swans)¶
Waterfowl have waterproof feathers with oil glands. Oil on the feather shaft may interfere with PCR — wipe the shaft gently with a clean tissue before placing in the envelope. Collect 2–3 flank feathers.

Why Paper Envelopes Are Mandatory¶
Feather follicles contain viable cells that need to breathe. Plastic bags trap residual moisture from the feather or environment, creating a microclimate that promotes mold and bacterial growth. Mold degrades DNA and renders the sample untestable. Paper envelopes allow air circulation while keeping the feathers contained.
